Analysis of Levocarnitine (NH2P-50 4E)

Levocarnitine is a hydrophilic substance used for the treatment of primary carnitine deficiency.
Levocarnitine was analyzed using Asahipak NH2P-50 4E, a polymer-based amino column, under HILIC mode condition. It was found that an addition of salt in the eluent stabilizes the retention of levocarnitine. Also, by introducing 50 mM KH2PO4 aq. solution as a preconditioning solvent helps shortening the column equilibration time.

[Note] This analysis requires column equilibration.
For column equilibration: Introduce 50 mM KH2PO4 aq. at 0.5 mL/min flow rate for 2 hours.
Then, change the solvent with the eluent.

Eluent       : 50 mM KH2PO4/CH3CN=35/65 (Adjusted with H3PO4 to a pH4.7)
Flow rate    : 1.0 mL/min
Detector     : UV (205 nm)
Column temp. : 30 °C
