LC/MS Analysis of Macrolide Antibiotics (C18U 2B)

The C18U series column is a silica-based reversed phase column especially designed for UHPLC use. The use of sub-2-micron organic silica hybrid particles provides superior separation efficiency and mechanical strength. In this application, macrolide antibiotics were analyzed simultaneously using an LC/MS/MS method with C18U 2B. For the analysis of basic drugs, ion-pair reagents are generally required with the use of an ordinary ODS column. However, C18U series columns have strong tolerance towards the alkaline condition. Thus, it can provide an ultra-rapid analysis of basic drugs using an alkaline condition, and without the use of ion-pair reagent.


Sample : 10 ng/mL each in H2O, 5 μL


Column       : Shodex C18U 2B (2.0 mm I.D. x 50 mm)
Eluent       : 0.05 % NH3 aq./CH3CN=40/60
Flow rate    : 0.4 mL/min
Detector     : ESI-MS (SIM)
