Effects of Instrument Settings on the Semi-micro Column Performance (KF-402HQ)

GPC KF-401HQ, KF-402HQ, and KF-403HQ are semi-micro type organic SEC (GPC) columns packed with 3 um gel. To achieve a best performance from a semi-micro column, it requires to optimize the LC system including detector cell volume, injection valve dead volume, system tubing, and detector response time.
This application shows the effects of detector cell volume on the separation of polystyrene oligomers using KF-402HQ. Fig.1 shows UV chromatograms and Fig.2 shows RI chromatograms. (A) and (C) are results using a semi-micro size detector cell while (B) and (D) are results using a regular (conventional) size detector cell. Both UV and RI detectors with regular detector cell showed dispersions of analytes in their chromatograms. This results from the use of large volume detector cell which led to the dispersions of analytes inside the cell. Therefore, it is recommended to use a semi-micro size detector cell (i.e., a small cell volume) to achieve the maximum column performance.
