Cleaning Method: Aqueous SEC (GFC) Columns

Please read the column cleaning method for your column.

For an efficient cleaning, reverse the flow direction.

Method 1: Adsorption of ionic (basic) compounds

Introduce the eluent with a salt concentration of 0.5 M or an acidic aqueous solution of pH 3. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 2: Adsorption of hydrophobic compounds

Introduce the eluent with 10 to 20 % (v/v) organic solvent (acetonitrile or methanol). Recommended solvent volume to introduce is 5 to 10 times of the column volume.

(Attention)
Proteins adsorbed on the column may precipitate by introducing an eluent containing an organic solvent.
 
Keeping the washing solution in the column for a long time will lead to column deterioration. Please replace the washing solution with the eluent immediately after cleaning.

OHpak SB-800 HQ series, OHpak SB-2000 series and OHpak LB-800 series

Method 1: Adsorption of hydrophobic compounds (when using aqueous eluent)

Introduce the eluent with a higher salt concentration. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 2: Adsorption of ionic compounds

Introduce the eluent with a high polar organic solvent (acetonitrile or methanol). Recommended solvent volume to introduce is 5 to 10 times of the column volume.

(Attention)
Keep the polar organic solvent and salt concentrations within the concentrations stated in the Operation Manual "List of Applicable Solvents".
 
Keeping the washing solution in the column for a long time will lead to column deterioration. Please replace the washing solution with the eluent immediately after cleaning.

UB series

For an efficient cleaning, reverse the flow direction. Set the flow rate at less than 0.3 mL/min.

 

Method 1: High salt concentration wash

Introduce the eluent with a salt concentration of 0.5 - 1 M. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 2: Organic solvent wash (when using aqueous eluent)

Introduce the eluent with 10 to 20 % (v/v) organic solvent (acetonitrile or methanol). Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 3: Alkaline solvent wash

Up to 1-M aqueous sodium hydroxide solution can be used for cleaning. Alternatively to wash off ionic components, add 1-M sodium chloride to the 1-M aqueous sodium hydroxide solution. Introduce the solvent of your choice for one hour.
If recovery was not observed, introduce the washing solvent for additional 5 to 10 times of the column volume.
