Cleaning Method: Polymer-based Columns for Reversed Phase Chromatography (RSpak)

Please read the column cleaning method for your column.

For an efficient cleaning, reverse the flow direction.

Method 1: Basic solvent wash (Apply after using the column under acidic to neutral condition for an extended period of time.)

Introduce 10-mM aqueous sodium hydroxide solution/acetonitrile = 80/20. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 2: Acid solvent wash (Apply after using the column under basic to neutral condition for an extended period of time.)

Introduce 50-mM aqueous phosphoric acid solution/acetonitrile = 90/10. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 3: Organic solvent wash (Apply after using the column under highly aqueous condition for an extended period of time.)

Introduce water/acetonitrile = 20/80. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

(Attention)

Keeping the washing solution in the column for a long time will lead to column deterioration. Please replace the washing solution with the eluent immediately after cleaning.
Method

For an efficient cleaning, reverse the flow direction.
Set the flow rate at 0.3 mL/min, and introduce 50 mL of methanol, acetonitrile or water. (Cleaning time: 165 minutes)

(Attention)
Keeping the washing solution in the column for a long time will lead to column deterioration. Please replace the washing solution with the eluent immediately after cleaning.

Method: adsorbing ionic componentsIntroduce the eluent and inject 20 - 50 μL of 2-M nitric acid aqueous solution several times.

Strong alkaline solvents such as 0.1-M aqueous sodium hydroxide solution can damage the detector cell. Do not connect the detector while regenerating the column and collect elute directly from the column outlet to the waste container.
