Cleaning Method: Cation Exchange Chromatography Columns

Please read the column cleaning method for your column.

For an efficient cleaning, reverse the flow direction.

 

Method

Set the flow rate at 0.5 mL/min. Introduce the eluent and inject 1 to 2 mL of 30 % (v/v) aqueous acetic acid solution several times.

Method 1: Adsorption of proteins

Introduce an eluent with higher salt concentration (about 1 M) or 0.1 % nonionic surfactants. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Method 2: Adsorption of hydrophobic compounds

Introduce a mixture of solvent containing 50-mM eluent (buffer or aqueous salt solution) and 10 to 20 % (v/v) acetonitrile. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

(Attention)
Keeping the washing solution in the column for a long time will lead to column deterioration. Please replace the washing solution with the eluent immediately after cleaning.

Introduce an eluent with higher salt concentration or 0.1-M aqueous acetic acid solution. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

 

Adsorption of hydrophobic compounds

Introduce a mixture of solvent containing 50-mM eluent (buffer or aqueous salt solution) and 50 % (v/v) acetonitrile or methanol. Recommended solvent volume to introduce is 5 to 10 times of the column volume.

(Attention)
Keeping the washing solution in the column for a long time will lead to column deterioration. Please replace the washing solution with the eluent immediately after cleaning.
