Title (626-23P) Analysis of Inorganic Anions using a New Hydroxide-selective Column
Abstract Ion chromatography is a well-established analytical technique used for the analysis of inorganic ions and small organic ions in aqueous samples. Hydroxide IC systems are advantageous in achieving lower background level than carbonate systems, especially with a suppressed current detector. We have developed a new IC column ShodexTM IC SI-36 4D compatible with hydroxide eluents, with improved selectivity for sulfite and sulfate. In addition to standard inorganic ions including; fluoride, chloride, nitrite, bromide, nitrate, sulfate and phosphate ions, sulfite ions can also be separated within 30min under isocratic elution mode. Additionally, basic features of the IC column ShodexTM IC SI-36 4D will be presented.

Title (627-20P) Direct Surfactant Analysis for Antibody Drugs using DAR for ADC with a Polymer-based Reverse Phase Column
Abstract Antibody drugs undergo various tests for quality control, including surfactant analysis and DAR (drug antibody ratio) analysis for ADC (antibody drug conjugate). We used a single column packed with polyhydroxymethacrylate-based spherical gel to achieve these two analyses. Quantification of surfactants in antibody drug formulation is generally difficult due to low UV absorption and protein or salt matrices. We developed a rapid HPLC method with this column using ELSD for quantifying polysorbate in a commercial antibody drug formulation, without the necessity of deproteinization nor desalting sample pretreatment. An ammonia - aqueous gradient and acetonitrile was used as eluent. Here, an alkaline eluent is effective for eluting IgG at void volume by size exclusion. Polysorbate can be retained by reverse phase, and quantified without interference from the matrices. This column can be also used for protein separation by hydrophobic chromatography. It is available for DAR analysis. When applied to the analysis of ADC (cysteine-linked type) with a gradient that reduced ammonium sulfate concentration, it was confirmed that a successful separation of DAR 0 to 6 was obtained. The reverse phase column packed the polyhydroxymethacrylate-based spherical gel is a powerful tool for surfactant and DAR analysis in quality control of antibody drugs.
