HILIC Mode Separation of Dextran 1 Oligomers (VG-25 4D)

“Dextran 1” is a low molecular weight fraction of dextran consisting of a mixture of isomaltooligosaccharides with an average molecular weight of 1,000. It is a widely used pharmaceutical ingredient as both active pharmaceutical ingredient and excipient.
In this application, Dextran 1 was analyzed by a polymer-based amino column, HILICpak VG-25 4D. Acetonitrile ratio in eluent was varied to examine its effects. All eluents achieved separations of glucose and over around 10 mer isomaltooligosaccharides. Among the eluents tested, eluent (2) provided a good separation balance across a wide range of glucose and isomaltooligosaccharides, making it the most suitable eluent composition for this sample.

Sample preparation

Weight 4 mg of Dextran 1 and completely dissolve it in 500 µL of water. After adding 500 µL of acetonitrile, filter through a membrane filter (0.45 μm) and use it as an injection sample.
