Effects of Sample Preparation Solution on Sugar Separation (VG-50 4D)

The effects of sample preparation solution composition on HILIC mode sugar separation were studied with a polymer-based amino column, HILICpak VG-50 4D. The eluent used was H2O/CH3CN = 20/80. It was observed that the more deviation of the sample preparation solution composition from that of the eluent, the worse the peak shape became, and theoretical plate number decreased. Sugars generally dissolve easily in water. However, to achieve a full separation performance, it is recommended to make the sample preparation solution composition as close as to the eluent composition. If a sample is difficult to dissolve in the chosen eluent, dissolve it in water first and then add acetonitrile to make the final acetonitrile concentration ratio to 50 % or more.
