Effects of Sample Injection Volume on Sugar Separation (VG-25 4D)

The effects of sample injection volume on HILIC mode sugar separation were studied with a polymer-based amino column, HILICpak VG-25 4D. The sample was dissolved in water and the sugar load was kept constant for all injection volumes. It was observed that the larger the injection volume, the worse the peak shape became, and theoretical plate number decreased. To achieve a full separation performance, it is recommended to make the sample preparation solution composition as close as to the eluent composition. If it is unavoidable to prepare a sample with a high-water ratio, it is recommended to reduce the injection volume as much as possible.
