Analysis of Cyanobacterial Glycogen Using SEC Column (SB-806M HQ)

Cyanobacterial glycogen has gained interest as a valuable biomass feedstock. In this application, glycogen extracts produced from Arthrospira platensis NIES-39 which is a kind of cyanobacteria were analyzed with OHpak SB-806M HQ, a column for SEC. The structural information including the type of sugar linkages in glycogen extracts is obtained by measuring the average MW of branching structure of glycogen after hydrolysis with isoamylase or glucoamylase.

(Black line) Native glycogen obtained from 10 mg dry weight
(Red line) Glycogen hydrolyzed with isoamylase
(Blue line) Glycogen hydrolyzed with glucoamylase

Columns      : Shodex OHpak SB-G 6B (6.0 mm I.D. x 50 mm) + SB-806M HQ (8.0 mm I.D. x 300 mm)
Eluent       : H2O
Flow rate    : 1.0 mL/min
Detector     : RI
Column temp. : 40 °C
